Gudrun Landgrebe was born on 20 June 1950 in Göttingen, Germany. She grew up in Bochum and attended theatre school in Cologne from 1968 to 1971. She made her stage debut at Stadttheater Bielefeld in 1971 and appeared as Klärchen Sisse in Heimat. Since 1981, she has frequently acted in German films, starting with the comedy Dabbel Trabbel. In 1983, she gained international recognition with the film Die flambierte Frau directed by Robert van Ackerens. She also appeared in Istvan Szabo's Oberst Redl (1985), Yerma (1984), Burkhard Driest's Annas Mutter (1984), and Liliana Cavani's The Berlin Affair (1985).
In 1997, Landgrebe acted in Rossini – oder die mörderische Frage, wer mit wem schlief, directed by Helmut Dietl and featuring Mario Adorf, Veronica Ferres, Heiner Lauterbach, Jan Josef Liefers, and Götz George. She appeared in the TV movie Opera Ball in 1998 alongside Heiner Lauterbach and Franka Potente. Since June 2001, she has been married to Dr. Ulrich von Nathusius and resides in Hunsrück.
